Lambda repressor binding to the OL1 consensus operator half site is accomplished by protein DNA interactions most notably between the helix-turn-helix motif and the N-terminal arm. The types of interactions that these sturctues have with DNA account for the recognition of lambda repressor operator sites. Mutations in key base pairs correlate with a drop in repressor affinity for operon sites.
All told, the lambda repressor's interactions with DNA have been well charactorized over the years thanks to the investigations via many groups. It remains the model system of choice in the discussion of cooporative binding at operator sites.
